Abstract

Las moléculas de HLA son moléculas presentadoras de péptidos, que muestran a los linfocitos T el estado de la célula y su entorno lo que permite activar la respuesta inmune específica en caso de infección o transformación tumoral. Existen dos tipos de moléculas de HLA, de clase I (HLA-A, -B y -C) y de clase II (HLA-DR, -DP y -DQ). Las primeras presentan péptidos procedentes del catabolismo de proteínas endógenas, mientras que las moléculas de HLA de clase II presentan péptidos exógenos, de membrana o de la ruta vesicular. La asociación genética más fuerte en muchas enfermedades autoinmunes es con varios alelos de HLA. Entre estas enfermedades están la artritis reumatoide, asociada a HLA-DR1, varios subtipos de DR4 y DR10, entre otras y no asociadas a otras moléculas como HLA-DR15. Por su parte, HLA-DR15 es un haplotipo asociado a esclerosis múltiple y expresa dos genes funcionales, HLA-DRB1*15:01 (DR2b) y HLA-DRB5*01:01 (DR2a). Por otro lado, se ha visto que varios péptidos pueden unirse a múltiples moléculas de HLA de clase II, lo que se ha denominado promiscuidad de las moléculas de HLA de clase II. Por otro lado, el proteasoma es un complejo multicatalítico abundante y es el principal sistema degradativo del citosol y el núcleo. Se han descrito tres tipos principales de proteasoma: el proteasoma constitutivo, con las subunidades catalíticas β1,β2 y β5; el inmunoproteasoma (β1i, β2i y β5i) y el timoproteasoma (β1i, β2i y β5t). Recientemente se han descrito proteasomas intermedios, conteniendo las subunidades del inmunoproteasoma β5i o β5i y β1i, junto con β1 y β2 o β2, respectivamente. En esta tesis se han abordado, mediante técnicas bioquímicas y de espectrometría de masas, varios aspectos del procesamiento y presentación antigénica. En primer lugar, se han analizado los repertorios peptídicos presentados por moléculas de HLA-DR asociadas diferencialmente a artritis reumatoide. En segundo lugar, se han comparado los repertorios peptídicos de las moléculas presentes en el haplotipo DR15: DR2a y DR2b. Finalmente se ha caracterizado la especificidad del proteasoma intermedio β5i y se ha estudiado su papel en la generación de ligandos peptídicos de HLA de clase I. Nuestros datos indican que entre los alotipos asociados a enfermedad, los repertorios peptídicos más similares son entre DR1 y DR10 y que un alto porcentaje de los péptidos comunes contien leucina en la posición P4 del core de unión y residuos básicos en la posicion P8. Por otro lado, los repertorios peptídicos de DR2a y DR2b ha permitido refinar los motivos de anclaje de dichas moléculas. Dichos repertorios son complementarios y poco solapantes. Además, se ha estimado que ambas moléculas aportan un número similar de péptidos al repertorio global de DR15. Un aspecto importante es que de todas las moléculas que hemos analizado, el grado de solapamiento de los repertorios peptídicos ha sido muy bajo en todas, lo que indica que, aunque la promiscuidad de HLA-DR existe, se produce en un bajo porcentaje de los ligandos peptídicos. Por otro lado, se ha desarrollado una nueva metodología para purificar proteasoma 20S activo y con alta pureza de forma rápida y eficiente. Los datos obtenidos indican que el proteasoma intermedio β5i presenta una actividad chymotrypsin-like menor que el proteasoma constitutivo, una actividad caspase-like similar, aunque un poco disminuida y una actividad tipo trypsin-like incrementada sobre el proteasoma constitutivo. Finalmente, se ha podido explicar, con digestiones in vitro de precursores peptídicos con proteasoma 20S purificado, la generación de varios ligandos de moléculas de HLA de clase I.


HLA molecules are peptide-presenting molecules, which show to T lymphocytes the state of the cell and environment, what allows to activate the specific immune response in case of infection or tumoral tranformation. There are two types of HLA molecules, class I (HLA-A, -B and -C) and class II (HLA-DR, -DP and -DQ). Class I molecules show peptides derived from the catabolism of endogenous proteins, while class II molecules show peptides derived from exogenous, membrane or vesicle proteins. The strong genetic association of many autoimmune diseases are with some HLA alleles. Among these disorders are rheumatoid arthritis, associated with DR1, several subtypes of DR4 and DR10, but not with DR15. HLA-DR15 is a complex haplotype associated with multiple sclerosis which expresses two functional genes, HLA-DRB1*15:01 (DR2b) and HLA-DRB5*01:01 (DR2a). On the other hand, several peptides can bind to multiple class II molecules, what has been denominated promiscuity of HLA class II molecules. On the other hand, the proteasome is an abundant multicatalytic complex which is the main degradative system in the cytosol and nucleus. Three major types of proteasome have been described: the constitutive proteasom with the catalytic subunits β1,β2 y β5; the immunoproteasome (β1i, β2i y β5i) and the thymoproteasome (β1i, β2i y β5t). Recently, intermediate proteasome have been described. These contain β1, β2 and β5i or β1i, β2 and β5i. In this thesis we have studied, using biochemical and mass spectrometry techniques, several points of the antigenic processing and presentation. First, the peptide repertoires bound to HLA-DR molecules differentially associated with rheumatoid arthritis have been analyzed. Second, we have studied the peptide repertoires associated to DR2a and DR2b, molecules simultaneously expressed in cells with the haplotype HLA-DR15. Finally, we have characterized the specificity of the intermediate proteasome β5i and its role in the generation of HLA class I peptide ligands. Our data indicate that, among the allotypes associated with the disease, the most similar peptide repertoires are DR1 and DR10, and many common peptides contain leucine in P4 core position and basic residues in P8. The peptide repertoires bound to DR2a and DR2b allowed the refinement of their binding motifs. Both repertoires are complimentary and with a low degree of overlap. Furthermore, we estimated that both molecules contribute similarly to the global peptide repertoire presented by HLA-DR15. The fact that all peptide repertoires analyzed showed a low degree or overlap indicate that, although the HLA-DR promiscuity exists, it occurs in a low number of peptide ligands. On the other hand, a new protocol for the rapid and efficient purification of an active and pure 20S proteasome has been developed. The data indicate that the intermediate proteasome β5i has a decreased chymotrypsin-like activity, a similar caspase-like and an increased trypsin-like activity than the standard proteasome. Finally, in vitro digestions with purified 20S proteasomes of peptide precursor of HLA class I-ligands has allowed to explain the generation of several HLA- class I peptide ligands.
