Abstract

A lo largo de la evolución, las plantas con flores han desarrollado un mecanismo genético de reconocimiento y rechazo específico del propio polen. Aunque desde el punto de vista evolutivo la autoincompatibilidad supone una ventaja, es un inconveniente en el cultivo de frutales, ya que puede limitar la producción. El almendro presenta autoincompatibilidad de tipo gametofítico, controlada por el locus S, que contiene al menos dos genes, expresados en el pistilo y en elpolen. El gen S del pistilo codifica para una glicoproteína con función ribonucleasa (ARNasa-S) que degrada específicamente el ARN del tubo polínico incompatible inhibiendo su crecimiento. El gen S del polen (SFB) codifica para una proteína con un dominio caja-F, que interviene en la degradación de proteínas mediante la vía del proteosoma 26S. Aunque la especificidad del rechazo del polen está controlada por el locus S, diversos estudios indican que existen genes modificadores no ligados a este locus que son indispensables en la vía bioquímica del sistema de incompatibilidad gametofítico. A pesar de que el almendro es una especie de origen autoincompatible, en la región italiana de Apulia se han identificado algunas variedades autocompatibles. En estas variedades el fenotipo autocompatible se asocia a la presencia de la ARNasa-Sf, sin embargo recientemente se han caracterizado almendros con el haplotipo Sf pero fenotipo autoincompatible lo que parece indicar la presencia de factores modificadores. El objetivo de esta Tesis Doctoral es avanzar en el conocimiento de los mecanismos moleculares de la incompatibilidad floral en almendro mediante la caracterización fenotípica, molecular, transcriptómica y proteómica de variedades y selecciones de almendro con haplotipo Sf y distinto fenotipo. Para ello se han empleado técnicas de secuenciación de ADN, que junto con técnicas que tienen como base la PCR han permitido conocer la secuencia de la región promotora de la ARNasa-Sf, de la ARNasa-Sf y de la región entre los genes ARNasa-Sf y SFBf. Por otro lado, también se han aplicado técnicas de retrotranscripción, de detección de actividad ribonucleasa y de microscopía de fluorescencia que nos han permitido observar el comportamiento de los elementos conocidos del sistema de incompatibilidad en almendro. Estos ensayos moleculares se han combinado a su vez con otros realizados en campo para caracterizar el fenotipo de las distintas variedades y selecciones utilizadas. Con el fin de caracterizar el perfil trancriptómico del sistema de incompatibilidad se han realizado análisis de RNAseq sobre pistilos polinizados y sin polinizar, confirmándose posteriormente los resultados mediante qPCR. La caracterización proteómica del sistema de incompatibilidad se realizó mediante iTRAQ de anteras, pistilos no polinizados y polinizados. Los resultados obtenidos de la caracterización fenotípica han mostrado una relación de incompatibilidad unilateral para el haplotipo Sf debido a una disfunción en el pistilo. Sin embargo, los ensayos de actividad ribonucleasa confirman que sólo están activas las ARNasas-Sf de las variedades incompatibles, aunque ambas ARNasas-Sf se transcriben. La secuenciación de ADN ha confirmado que las secuencias de las ARNasas-Sf y de sus regiones promotoras son iguales. Sólo se ha observado la insercción de timinas en la región intergénica, que podrían ser responsables de la transcripción diferencial de un ORF descrito por primera vez en esta zona. Los análisis de transcriptómica y proteómica han ofrecido una visión general de todos elementos involucrados en las reacciones de incompatibilidad y han mostrado varios candidatos como posibles factores modificadores del sisitema de incompatibilidad.


Over the evolution, the plants with flowers have developed a genetic mechanism of specific recognition and rejection of self pollen. Although from the evolutionary point of view self-incompatibility is an advantage, for fruit trees it may be a disadvantage for, since it limits their production. Almond presents gametophytic self-incompatibility controlled by the S locus, which at least contains two genes. The pistil S gene encodes a glycoprotein with ribonuclease function (S-RNase), which specifically degrades the RNA of the incompatible pollen and inhibits its growth. The pollen S gene (SFB) codes for an F-box protein involved in the degradation of proteins by the 26S proteosome. Although the specificity of pollen rejection is controlled by the S locus, several studies indicate that modifier genes not linked to this locus are involved in the biochemical pathway of the gametophytic incompatibility system. Despite the almond is a self-incompatible species, in the Italian region of Apulia some self-compatible cultivars have been identified. In these cultivars the self-compatible phenotype is associated with the presence of the Sf RNase, however, almonds with the Sf haplotype and a self-incompatible phenotype have recently been characterized, what seems to indicate the presence of modifier factors. Therefore, the objective of this PhD Thesis is to advance in the knowledge of the molecular mechanisms of floral incompatibility in almond through the phenotypic, molecular, transcriptomic and proteomic characterization of almond cultivars and selections with the Sf haplotype but a different phenotype. For this, DNA sequencing techniques have been used, which together with PCR-based techniques allowed to know the sequence of the promoter region of the Sf-RNase, the Sf-RNase and the region between Sf-RNase and SFBf genes. On the other hand, retrotranscription, detection of ribonucleases activity and fluorescence microscopy techniques have also been applied, which allowed to observe the behavior of the known elements of the self-incompatibility system in almond. These molecular assays have been combined with field assays to characterize the phenotype of the different almond selections and cultivars used. In order to characterize the transcriptomic profile of the incompatibility system, RNAseq analyzes were performed on pollinated and non-pollinated pistils. The results are then confirmed by qPCRs. The proteomic characterization of the incompatibility system was performed by iTRAQ of anthers, non-pollinated and pollinated pistils. The results obtained from the phenotypic characterization have shown a unilateral incompatibility relation for the Sf haplotype due to a dysfunction in the pistil. However, ribonuclease activity assays confirmed that only the Sf-RNase of the incompatible selections were active, although both Sf-RNases were transcribed. DNA sequencing confirmed that the sequences of the Sf-RNase and those of their promoters were identical. Only thymines insertion of three into the intergenic region was observed, which could be the responsible for the differential transcription of an ORF, firstly described in this region. Transcriptomic and proteomic analyzes have provided an overview of all elements involved in incompatibility reactions and have shown some candidates as putative incompatiblity system modifier factors.
